Premature ovarian failure (POF) is a very common disease. Directly inhibiting the activity of mammalian target of rapamycin (mTOR) can treat POF, but it can also have adverse effects on women. Professor Shi Zhonghua, Professor Zhao Chun, Professor Wang Fuqiang and Professor Liang Gaolin from Nanjing Medical University co-assembled hydrogel factor Y and receptor tyrosine kinase (RTK, the upstream kinase of mTOR) inhibitor Inh to form a hydrogel Glue Y+Inh, and use it to achieve intelligent, RTK-responsive Inh release.



Key points of this article:

(1) The released Inh can moderate mTOR activity, thereby delaying ovarian senescence. Experimental results show that gel Y+Inh can promote the meiotic maturation of oocytes and the early embryonic development of fertilized eggs.




(2) In vivo animal experiments show that gel Y+Inh can stimulate the ovaries to secrete estrogen and progesterone by down-regulating mTOR activity, thereby promoting the development of more antral follicles and effectively delaying ovarian aging in elderly mice. In summary, this new type of hydrogel Y+Inh is expected to achieve clinical treatment of POF in the near future and delay ovarian aging.
