Anti-inflammatory colchicine therapy has ushered in a new era in the treatment of atherosclerotic cardiovascular disease, but its clinical benefits are not yet fully clear. This study proposes a dual-coordination-driven strategy, successfully constructing stable colchicine metal-organic nanostructures (COL-TA-Zn) by integrating the tropolone ring of colchicine (COL), the phenolic hydroxyl groups of tannic acid (TA), and zinc ions (Zn²⁺). This design combines the antioxidant and anti-inflammatory properties of COL and TA,Construct a nanoplatform capable of scavenging free radicals and regulating inflammatory pathways. Based on the strong coordination effect of Zn²⁺, the resulting COL-TA-Zn nanocomposite exhibits enhanced stability under physiological conditions, ensuring efficient delivery and sustained bioactivity. In vitro experiments confirmed that it can inhibit foam cell formation and the release of various inflammatory mediators, indicating that this system simultaneously targets oxidative stress and inflammation.It has significant potential in the prevention and treatment of atherosclerosis. In the Apoe−/− mouse model, intravenous injection of COL-TA-Zn significantly reduced the area of atherosclerotic plaques and decreased the levels of matrix metalloproteinase-9 (MMP-9), tumor necrosis factor-α (TNF-α), and reactive oxygen species (ROS). Its anti-atherosclerotic efficacy is notably superior to treatment with colchicine alone. These findings highlight the potential application of the dual-coordination-driven nanoplatform in cardiovascular disease therapy.
