This research, reported by Professor Wang Yiguang from Peking University, Professor Zhang Qiang, and Associate Researcher Chen Binlong, can be summarized from the perspective of biomaterials as follows: This work proposes a "in situ cancer vaccine" strategy. The authors constructed a nano-light-sensitive adjuvant system that can be injected intravenously. Through the nanoparticle endocytosis localization triggered by the tumor microenvironment, it produces reactive oxygen species under light conditions, inducing GSDME-mediated pyroptosis in tumor cells, thereby releasing a large amount of tumor antigens. At the same time, it simultaneously releases immune adjuvants to activate the innate immune pathway, promoting dendritic cell antigen presentation and T-cell immune responses. Through this "material regulation of tumor cell death mode → converting tumors into sources of their own vaccines" strategy, this system significantly enhanced systemic anti-tumor immunity and inhibited distant tumor growth in mouse models, demonstrating the potential of biomaterials in precisely regulating the tumor immune microenvironment and constructing individualized cancer vaccines. The related research, titled "In situ-generated vaccine-like pyroptosome for personalized cancer immunotherapy", was published in Nature Materials.
