Advanced Materials | A bio-lubricating nanoplatform based on allicin, for reshaping the inflammation-aging axis in periodontitis treatment

已传文件:photo/1773121782.png Periodontitis is driven by a self-reinforcing cycle of persistent inflammation and cellular senescence, and the colonization of pathogenic microorganisms further exacerbates the situation. To address this challenge, inspired by the "fortress effect", we report a nanobiological lubrication platform based on allicin (PPCG), which establishes physical and biological protective barriers to precisely regulate the periodontal microenvironment. PPCG integrates the hydrated lubricating bimodal copolymer P (DMA-bMPC) (PDMPC) with bioactive allicin. The lubricated PDMPC barrier may inhibit the adhesion of pathogenic microorganisms and the formation of biofilms, forming an "outer fortress" to resist bacterial invasion. At the same time, the continuous release of allicin can regulate bone marrow mesenchymal stem cells (BMMSC) and alleviate the inflammatory response. It can retain stemness and pluripotent differentiation potential, thereby forming an "internal defense fortress" to promote the regeneration of soft and hard tissues. This dual protective barrier can significantly slow down the aging and inflammation of periodontal tissues and prevent alveolar bone loss in mouse periodontitis models. Moreover, PPCG can also rebalance the oral microbiota and maintain ecological stability. The therapeutic effect has also been confirmed through an artificial intelligence-assisted detection system based on the YOLO v8 deep learning model. Overall, this study proposes a therapeutic intervention strategy for periodontitis, providing an expandable and transformable treatment method for inflammation and aging.
