Delayed endothelialization and inflammatory responses are two major obstacles that drug-eluting stents (DES) face in promoting vascular healing. This paper reports a biomimetic nanozyme strategy to tackle this issue. First, we designed a stable glutathione peroxidase (GPx) mimicking nanozyme with a diselenide-polyphenol network structure and anchored it onto the stent. Then, using its reversible interactions with polyphenols, we efficiently loaded the thrombin inhibitor bivalirudin onto the nanozyme. This nanozyme has strong GPx activity, can controllably generate nitric oxide (NO), and release bivalirudin on demand. The synergistic effect of NO and bivalirudin effectively inhibits thrombin activity, prevents platelet activation, and thereby reduces thrombosis and related inflammatory responses. In vitro pathological model cell studies show that the nanozyme-engineered stent can specifically inhibit smooth muscle cell (SMC) proliferation, regulate macrophage polarization, and promote repair and proliferation of endothelial cells. As a result, this stent can promote rapid endothelialization and vascular healing, preventing restenosis in rabbit and pig models.
