I. Overview
Amino activated agarose magnetic beads (Magarose-NH2) is a kind of 6% cross-linked magnetic agarose microspheres. The magnetic beads contain reactive primary amines at the end of the Linked for affinity purification. Gels are ideal for immobilizing peptides and are used for affinity purification of antibodies or other binding partners.
Although other amine-reactive methods can immobilize molecules on amino-activated agarose beads, carbodiimide (EDC) is unique among cross-linking agents, enabling coupling via -COOH. In a near-neutral to acidic pH buffer, EDC can effectively react with carboxylates to form intermediate esters, which react with nucleophiles such as primary amines on amino-activated sepharose beads. In most affinity purification methods, the amide bond between the molecule and the agarose gel carrier is stable and can prevent leakage.
Amino activated agarose magnetic beads are also suitable for immobilizing oligonucleotides via 5-phosphate groups. In the presence of imidazole, EDC effectively reacts with phosphoric acid groups in approximately the same way as carboxylates.
