Photodynamic therapy (PDT) is a non-invasive treatment strategy for cancer treatment, but it always suffers from the low reactive oxygen species (ROS) yield of traditional organic dyes.
In view of this, Tang Benzhong, Academician of Hong Kong University of Science and Technology, Jiang Xingyu of Southern University of Science and Technology, and Zheng Wenfu of National Nanoscience Center, and others introduced a lipid-encapsulated aggregation-induced luminescence nanoparticles (AIE NPs) under near-infrared light (800 nm). It has a high quantum yield (23%) and a maximum two-photon absorption (TPA) cross section of 560 GM.
Key points of this article:
1) On mouse melanoma models, AIE-NPs can be used as imaging agents for spatiotemporal imaging of tumor tissues, with a penetration depth of up to 505μm. NIR-1 (800 nm) two-photon fluorescence imaging showed that AIE NPs accumulated in the tumor tissues of the mouse melanoma model, especially the tumor blood vessels.
2) Importantly, AIE-NPs can simultaneously produce singlet oxygen (1O2) and highly toxic hydroxyl radicals (•OH) under the irradiation of 800nm photodynamic tumor ablation. In addition, after imaging and treatment, AIE-NPs can be effectively eliminated from mice.
In summary, this research provides a strategy for developing tumor image-guided PDT therapeutics with high brightness, high light stability and high biological safety.
