Excess zinc ions (Zn2+) in living cells have been identified as a tumor biomarker for early cancer diagnosis and cancer progression monitoring. Xuzhou Medical University Hengliang Shi and Associate Professor Gao Fenglei developed a new type of carbon nanohorn/platinum nanoparticle/DNA (CNHs/Pt NPs/DNA) nanoplatform through clamp hybridization chain reaction (c-HCR) process, and It is used to image Zn2+ in cancer cells and enhance collaborative optical therapy.
Key points of this article:
(1) In the experiment, the cross-type DNAzyme (c-DNAzyme), hairpin DNA1, hairpin DNA2 and aptamer DNA are adsorbed on the surface of CNHs/Pt NPs, and the fluorescence of carboxytetramethylrhodamine will be quenched at this time. After entering the living cells, c-DNAzyme will be lysed, and in the presence of Zn2+ in the cell, it will output trigger DNA, and then start the c-HCR process for fluorescence amplification.
(2) Compared with a single HCR process triggered by a single DNAzyme, the c-HCR process can further improve the efficiency and sensitivity of amplification. In addition, the nanoprobe also has catalytically enhanced photodynamic and photothermal effects, which proves that it is an efficient platform for cancer diagnosis and treatment.
