Artificial peroxidase has attracted much attention due to its practicability in the construction of protocell system and its great potential in the treatment of diseases. However, it is still a major challenge to prepare a viable artificial peroxidase under physiological conditions to supplement multiple stable functions. Here, Academicians Ushen, Fan Kelong, Yan Xiyun and others of Institute of Biophysics, Chinese Academy of Sciences reported a new strategy to design an artificial peroxidase, which uses nanase to regulate the activity of various similar enzymes to adapt to the enzymatic activity of natural peroxidase.
Main points of this article:
1) By doping iron and nitrogen, the active site of the enzyme was introduced into the graphitized part of the hollow carbon nano shell to form the Fe-N4 coordination and atomic Fe cluster. With Fe cluster as reversible cofactor and FE-N4 as cogroup, the carbon nanase obtained showed stable and multiple peroxidase-like activities, including catalase, uricase, superoxide dismutase, peroxidase and oxidase, which were called pero-nanozysome.
2) This artificial peroxidase has therapeutic effects on hyperuricemia and protecting neurons from free radicals generated during ischemic stroke by using uricase-catalase and superoxide dismutase-catalase in tandem, respectively.
In summary, Pero-nanozysome is a promising candidate for the design of artificial peroxidase with in vivo function.
