How to make nanoparticles evade immune surveillance and penetrate tumor tissue deeply is of great significance for improving the therapeutic effect of nanomedicine. Professor Shaobing Zhou and Professor Jingya Zhao from Southwest Jiaotong University have developed an extracellular vesicle nanoplatform that responds to near-infrared (NIR) light. It has long blood circulation, can penetrate deeply into tumor tissues, and is quickly cleared after treatment. And other advantages.






Key points of this article:

(1) Similar to the design of the Trojan horse, the experiment “hidden” the anti-tumor drug DOX and 4.2 nm Ag2S quantum dots into vesicles secreted by macrophages through electroporation to prepare the nano-platform. The natural components of extracellular vesicles and tumor-targeting activity enable the nanoplatform to achieve high aggregation in tumors, and its biological distribution in vivo can be studied through near-infrared fluorescence imaging generated by Ag2S quantum dots.






(2) After the nanomedicine is accumulated in the tumor site, the experiment uses the near-infrared photothermal effect of Ag2S quantum dots to release the drug from the "Trojan Horse". At the same time, the released ultra-small quantum dots and DOX can penetrate the entire tumor with a diameter of about 9 mm, thereby effectively killing tumor cells. In addition, inorganic Ag2S quantum dots can be cleared by the kidneys after treatment to be quickly excreted from the body, thereby avoiding potential toxic side effects.
