Fenton reaction (pH 3~4) has harsh reaction conditions and slow reaction speed, which severely restricts the efficacy of chemokinetic therapy (CDT). Here, Zhao Yanli and others from Nanyang Technological University in Singapore reported a super-small three-metal (Pd, Cu and Fe) alloy nanoenzyme (PCF-a NEs), which has a dynamic active site synergy, so it is close to neutral. It exhibits cascade mimetic activity of glutathione peroxidase and peroxidase (POD) at pH.
Key points of this article:
1) PCF-a NEs has a photothermal enhanced POD characteristic, and has a high photothermal conversion efficiency (62%), which is used to coordinate tumor cell apoptosis.
2) In addition, ultrasound can also enhance the mass transfer at the active catalytic sites of PCF-a NEs, thereby accelerating the Fenton-like reaction of tumor-specific CDT.
This work provides a strategy to construct alloy nanoenzymes in a biologically inspired way to amplify active oxygen in tumors in response to external stimuli, thereby showing higher tumor growth inhibition efficiency in vitro and in vivo.
