The effectiveness of immune checkpoint blockade (ICB) therapy will be affected by the immunosuppressive tumor microenvironment (TME). In clinical tumor treatment, how to reverse the immunosuppressive state is still a challenge that needs to be solved urgently. Immunogenic cell death (ICD) is a method of inducing the immune system of therapeutic tumors.
In view of this, researcher Lin Jun and researcher Cheng Ziyong of Changchun Institute of Applied Chemistry, Chinese Academy of Sciences have constructed CO2-g-C3N4-Au@ZIF-8@F127(CCAZF) photocatalytic nanogenerator, which can treat carbon monoxide (CO) gas and Immunotherapy is integrated to overcome the limitations of monotherapy, thereby improving anti-tumor immunity, anti-metastasis and preventing recurrence.
The main points of this article: (1) CCAZF has high-efficiency light-controlled release of CO performance, which can gradually aggravate the oxidative stress in tumor cells to achieve ICD. Under the induction of ICD, CO therapy can enhance the immune response and can effectively activate immune cells.
2) When combined with ICB, CCAZF can show enhanced immunity, and then achieve the regression of primary and remote tumors. In summary, this in-situ photocatalytic CO treatment strategy can avoid the toxic and side effects of CO to the greatest extent, thereby providing a new method for designing new ICD inducers.
