Nanomaterials and the immunological behavior of the immune system provide a theoretical basis for the development of immunotherapy methods based on nano-vaccine. Here, Professor Li Yanmei of Tsinghua University and researcher Yu Xuefeng of the Shenzhen Institute of Advanced Technology of the Chinese Academy of Sciences systematically studied the immune enhancement effects of black phosphorus nanosheets (BPs) in vivo and in vitro, and proposed a new and effective multifunctional nano adjuvant.
In this paper, the phenylalanine-lysine-phenylalanine (FKF) tripeptide modified epitope was used to simply coat BPs to prepare the FKF-OVAp@BP nano-vaccine, which has high loading capacity and high efficiency Drug delivery, ability to activate dendritic cells (DC) and good biocompatibility, can be used for cancer immunotherapy. In a mouse model, the nano-vaccine can induce effective anti-tumor cell immunity through systemic immunization, and significantly enhance the checkpoint blockade (CPB) therapeutic effect on melanoma. In addition, the near-infrared (NIR) photothermal effect of BPs can also provide a favorable immune microenvironment for improving local immunity. In summary, this research provides a new strategy for the use of nano-adjuvants to combine immune activity and photothermal effect to enhance cancer immunotherapy, and is expected to further promote the development and application of multifunctional auxiliary nanomaterials for cancer treatment .
