Summary:
This study designed a MgAl-LDH@N/GP composite hydrogel, introducing NGF-loaded MgAl-LDH nanosheets into a GelMA/PDA system. The hydrogel is injectable and can be cross-linked in situ with light, activating the TRPV1 channels of sensory neurons under mild photothermal conditions. This promotes SP secretion, reverses the phenotype of inflammatory osteoclast precursors, and continuously releases NGF and Mg²⁺. The research shows that it improves the neuro-immune microenvironment, promotes vascular-osteogenesis coupling, and aids in repairing osteoporotic bone defects. The mechanism involves SP regulating ATF4-CHAC1-related oxidative stress and inflammatory phenotypes.
The hydrogel has a suitable pore structure, degradability, and photothermal stability, and can slowly release NGF and Mg²⁺, with good biocompatibility. Mild photothermal treatment promotes calcium influx and SP secretion in DRG, enhancing axon growth. SP-conditioned medium reduces TRAP-positive multinucleated cells and CX3CR1 expression, downregulates Itgam, IL-23a, Tnf, and IL-1b, and upregulates Tgfb1 and IL-10; RNA-seq suggests involvement of ATF4-CHAC1, oxidative phosphorylation, and proteasome pathways. The conditioned medium promotes BMSC RUNX2, OPN, ALP, and mineralization, and enhances tube formation in HUVECs. In vivo, it promotes β3-tubulin/SP neural reconstruction, reduces TRAP⁺CX3CR1⁺ inflammatory osteoclast precursors, and boosts CD31, VEGF, RUNX2, OCN, and bone repair.
